As in ; do you / can you use some of your current lenses with the new body , or are you on a 'free for all ' change? I'm desperately wanting a canon 5D mkII, as i have a canon 400D and would like to stay with the same brand.

Im a canon girl... but I know nikon is great too... It all really depends on your intentions I suppose... but I would say start medium grade if you intend on eventually upgrading... but if you don't intend on upgrading, A Canon Rebel XTi is a GREAT first DSLR.
